Alfrun Kliems hat Russistik und Bohemistik in Berlin und Prag studiert und 2000 über im Exil deutsch schreibende tschechische Schriftsteller_innen promoviert. Sie hat lange als Fachkoordinatorin für die Literaturen Ostmitteleuropas am Leipziger GWZO geforscht und wurde 2011 als Professorin für Westslawische Literaturen an die Humboldt-Universität Berlin berufen. Sie beschäftigt sich mit Fragen von Migration und Sprachwechsel in der Literatur, dem Underground in Ostmitteleuropa sowie polnischen, tschechischen und slowakischen Comics über die Romantik.

She has conducted research as the coordinator of the department for Eastern Central European Literature at the GWZO in Leipzig and was appointed professor for Western Slavic Literature at the Humboldt University Berlin in 2011.
